چکیده
Androgenetic alopecia is a common form of pattern hair loss, characterized by miniaturized follicles (HFs) at the front and parietal scalp, while hairs on occipital scalp are preserved. Moreover, different sites human body exhibit distinct types patterns HFs. Understanding molecular basis this heterogeneity will help to design targeted treatment strategies. The Wnt signalling pathway its Dickkopf (Dkk) inhibitors have been suggested regulate HF induction patterning. We previously shown that Dkk4 specifically expressed in placodes during mouse skin development. To elucidate functions patterning, work, we generated Dkk4-knockout mice using CRISPR/Cas9. mutants showed disrupted patterning with an increase interplacodal distance. Surprisingly, lateral back completely lacked first wave In order address regional differences formation, Dkk4-EGFP knockin line, which recapitulated expression placodes. Our data revealed intra- Dkk4-EGFP, suggesting WNT-DKK axis underlies specificities Elucidating mechanisms shed light loss such as androgenetic alopecia.

Miz1 is required for hair follicle structure and hair morphogenesis.
Previous work has implicated the Myc-binding transcription factor Miz1 in the control of keratinocyte proliferation and in the cellular response to TGFbeta. Miz1 is expressed in basal keratinocytes of the interfollicular epidermis and in hair follicles. Here we have conditionally knocked out the POZ/BTB transactivation domain of Miz1 in keratinocytes using a keratin 14 (K14)-Cre mouse deleter s...
متن کاملEpithelial Wnt ligand secretion is required for adult hair follicle growth and regeneration
β-Catenin, a key transducer molecule of Wnt signaling, is required for adult hair follicle growth and regeneration. However, the cellular source of Wnt ligands required for Wnt/β-catenin activation during anagen induction is unknown. In this study, we genetically deleted Wntless (Wls), a gene required for Wnt ligand secretion by Wnt-producing cells, specifically in the hair follicle epithelium ...
متن کاملNoggin is required for induction of the hair follicle growth phase in postnatal skin.
During postnatal development, the hair follicle (HF) shows cyclic activity with periods of relative resting, active growth (anagen), and regression. We demonstrate that similar to the HF induction in embryonic skin, initiation of a new hair growth phase in postnatal skin requires neutralization of the inhibitory activity of bone morphogenetic protein 4 (BMP4) by the BMP antagonist noggin. In th...
متن کاملHierarchical patterning modes orchestrate hair follicle morphogenesis
Two theories address the origin of repeating patterns, such as hair follicles, limb digits, and intestinal villi, during development. The Turing reaction-diffusion system posits that interacting diffusible signals produced by static cells first define a prepattern that then induces cell rearrangements to produce an anatomical structure. The second theory, that of mesenchymal self-organisation, ...
متن کاملWnt Signaling in Hair Follicle Development
HF is a mini-organ formed with mesenchymal-epithelial interaction (Schmidt-Ullrich & Paus, 2005). HF development and cycle are depended on signaling between its epidermal and dermal components. Especially, dermal organizing center, DP, is both chemical and physical niche for epithelial stem cells. DP plays an important role in HF induction and cycling by controlling self-renewal and differentia...
